                  <content styleCode="italics">Clostridium difficile</content> associated diarrhea (CDAD) has been reported with nearly all antibacterial agents, including minocycline, and may range in severity from mild diarrhea to fatal colitis. Treatment with antibacterial agents alters the normal flora of the colon leading to overgrowth of <content styleCode="italics">C. difficile</content>.</paragraph>
                <paragraph>
                  <content styleCode="italics">C. difficile</content> produces toxins A and B which contribute to the development of CDAD. Hypertoxin producing strains of <content styleCode="italics">C. difficile</content> cause increased morbidity and mortality, as these infections can be refractory to antimicrobial therapy and may require colectomy. CDAD must be considered in all patients who present with diarrhea following antibiotic use. Careful medical history is necessary since CDAD has been reported to occur over two months after the administration of antibacterial agents.</paragraph>
                <paragraph>If CDAD is suspected or confirmed, antibiotic use not directed against <content styleCode="italics">C. difficile</content> may need to be discontinued. Appropriate fluid and electrolyte management, protein supplementation, antibiotic treatment of <content styleCode="italics">C. difficile</content>, and surgical evaluation should be instituted as clinically indicated.</paragraph>

Under such conditions, lower than usual total doses are indicated, and if therapy is prolonged, serum level determinations of the drug may be advisable.</paragraph>

Clinical manifestations of intracranial hypertension include headache, blurred vision, diplopia and vision loss; papilledema can be found on fundoscopy. Women of childbearing age who are overweight or have a history of IH are at a  greater risk for developing intracranial hypertension. Concomitant use of isotretinoin and tetracycline should be avoided because isotretinoin, a systemic retinoid, is also known to cause intracranial hypertension.</paragraph>
                <paragraph>Although intracranial hypertension typically resolves after discontinuation of treatment, the possibility for permanent visual loss exists. If visual disturbance occurs during treatment, prompt ophthalmologic evaluation is warranted. Because intracranial pressure can remain elevated for weeks after drug cessation, patients should be monitored until they stabilize.</paragraph>

Sporadic cases of serum sickness have presented shortly after minocycline use. Symptoms may be manifested by fever, rash, arthralgia, and malaise. In symptomatic patients, immediately discontinue the use of all tetracycline-class drugs, including MINOLIRA.</paragraph>

Fever and lymphadenopathy may be present. In some cases, death has been reported. If this syndrome is recognized, discontinue MINOLIRA immediately.</paragraph>

In this study, body weight gain was significantly reduced in pregnant females that received 50 mg/kg/day (resulting in approximately 2.5 times the systemic exposure to minocycline observed in patients administered MINOLIRA). No effects of treatment on the duration of the gestation period or the number of live pups born per litter were observed. Gross external anomalies observed in F1 pups (offspring of animals that received minocycline) included reduced body size, improperly rotated forelimbs, and reduced size of extremites.                 <paragraph>In a carcinogenicity study in which minocycline hydrochloride was orally administered to male and female rats once daily for up to 104 weeks at dosages up to 200 mg/kg/day, minocycline hydrochloride was associated in both genders with follicular cell tumors of the thyroid gland, including increased incidences of adenomas, carcinomas, and the combined incidence of adenomas and carcinomas in males, and adenomas and the combined incidence of adenomas and carcinomas in females. In a carcinogenicity study in which minocycline hydrochloride was orally administered to male and female mice once daily for up to 104 weeks at dosages up to 150 mg/kg/day, exposure to minocycline hydrochloride did not result in a significantly increased incidence of neoplasms in either males or females.  </paragraph>
                <paragraph>Minocycline was not mutagenic in vitro in a bacterial reverse mutation assay (Ames test) or CHO/HGPRT mammalian cell assay in the presence or absence of metabolic activation. Minocycline was not clastogenic in vitro using human peripheral blood lymphocytes or in vivo in a mouse micronucleus test.</paragraph>
                <paragraph>Male and female reproductive performance in rats was unaffected by oral doses of minocycline of up to 300 mg/kg/day (which resulted in up to approximately 40 times the level of systemic exposure to minocycline observed in patients administered MINOLIRA). However, oral administration of 100 or 300 mg/kg/day of minocycline to male rats (resulting in approximately 15 to 40 times the level of systemic exposure to minocycline observed in patients administered MINOLIRA) adversely affected spermatogenesis. Effects observed at 300 mg/kg/day included a reduced number of sperm cells per gram of epididymis, an apparent reduction in the percentage of sperm that were motile, and (at 100 and 300 mg/kg/day) increased numbers of morphologically abnormal sperm cells. Morphological abnormalities observed in sperm samples included absent heads, misshapen heads, and abnormal flagella.   </paragraph>

            <paragraph>The safety and efficacy of minocycline hydrochloride extended-release tablets in the treatment of inflammatory lesions of non-nodular moderate to  severe acne vulgaris was assessed in two 12-week, multi-center, randomized, double-blind, placebo-controlled, trials in subjects ≥12 years. The mean age of subjects was 20 years and subjects were from the following racial groups: White (73%), Hispanic (13%), Black (11%), Asian/Pacific Islander (2%), and Other (2%). </paragraph>
            <paragraph>In two efficacy and safety trials, a total of 924 subjects with non-nodular moderate to severe acne vulgaris received minocycline hydrochloride extended-release tablets (approximately 1 mg/kg) or placebo for a total of 12 weeks.</paragraph>
            <paragraph>The two primary efficacy endpoints were:</paragraph>
            <paragraph>1) Mean percent change in inflammatory lesion counts from Baseline to 12 weeks.</paragraph>
            <paragraph>2) Percentage of subjects with an Evaluator's Global Severity Assessment (EGSA) of clear or almost clear at 12 weeks.</paragraph>
            <paragraph>Efficacy results are presented in Table 5.</paragraph>

              <content styleCode="bold">WHAT IS MINOLIRA?</content>
            </paragraph>
            <paragraph>MINOLIRA is prescription medicine used to treat only pimples and red bumps (non-nodular inflammatory lesions) that happen with moderate to severe acne vulgaris in people 12 years and older.</paragraph>
            <paragraph>MINOLIRA should not be used for the treatment of infections.</paragraph>
            <paragraph>It is not known if MINOLIRA is safe and effective in children under the age of 12 years.</paragraph>
            <paragraph>
              <content styleCode="bold">Who should not take MINOLIRA?</content>
            </paragraph>
            <list listType="unordered" styleCode="disc">
              <item>Do not take MINOLIRA if you are allergic to medicines in the tetracycline class. Ask your healthcare provider or pharmacist for a list of these medicines if you are not sure.</item>
            </list>
            <paragraph>
              <content styleCode="bold">Before you take MINOLIRA, tell your healthcare provider about all of your medical conditions, including if you:</content>
            </paragraph>
            <list listType="unordered" styleCode="disc">
              <item>have kidney problems</item>
              <item>have liver problems</item>
              <item>have diarrhea or watery stools</item>
              <item>have vision problems.</item>
              <item>are pregnant or plan to become pregnant. MINOLIRA may harm your unborn baby. Taking MINOLIRA while you are pregnant may cause serious side effects on the growth of bone and teeth of your baby. Stop taking MINOLIRA and call your healthcare provider right away if you become pregnant during treatment with MINOLIRA.</item>
              <item>are breastfeeding or plan to breastfeed. MINOLIRA can pass into your breast milk and may harm your baby. You should not breastfeed during treatment with MINOLIRA.</item>

              <content styleCode="bold">Tell your healthcare provider about all the other medicines you take,</content> including prescription and over-the-counter medicines, vitamins, and herbal supplements. MINOLIRA and other medicines may affect each other causing serious side effects.</paragraph>
            <paragraph>Especially tell your healthcare provider if you take:</paragraph>

              <item>birth control pills. MINOLIRA may reduce the effectiveness of certain birth control pills. You could become pregnant. You should use a second form of birth control during treatment with MINOLIRA. Talk to your healthcare provider about what types of birth control you can use to prevent pregnancy during treatment with MINOLIRA.</item>
              <item>a medicine taken by mouth that contains isotretinoin.</item>

                <content styleCode="bold">Do not miss your dose of MINOLIRA.</content> Missing doses or not taking all doses of MINOLIRA may: 							<list listType="unordered" styleCode="circle">
                  <item>make the treatment not work as well</item>
                  <item>increase the chance that the bacteria will become resistant to MINOLIRA</item>
                </list>
              </item>
              <item>Take MINOLIRA with or without food. Taking MINOLIRA with food may lower your risk of getting irritation or ulcers in your esophagus. Your esophagus is the tube that connects your mouth to your stomach.</item>
              <item>Do not crush or chew MINOLIRA tablets.</item>
              <item>If you take too much MINOLIRA, call your healthcare provider right away or go to the nearest hospital emergency room.</item>
            </list>
            <paragraph>
              <content styleCode="bold">What should I avoid during treatment with MINOLIRA?</content>
            </paragraph>
            <list listType="unordered" styleCode="disc">
              <item>Avoid sunlight or artificial sunlight, such as sunlamps or tanning beds. You could get severe sunburn. Use sunscreen and wear loose-fitting clothes that cover your skin while out in sunlight. Stop taking MINOLIRA if you get sunburn.</item>
              <item>You should not drive or operate dangerous machinery until you know how MINOLIRA affects you. MINOLIRA may cause you to feel dizzy or lightheaded, or have a spinning feeling (vertigo).</item>
            </list>
            <paragraph>
              <content styleCode="bold">What are possible side effects of MINOLIRA?</content>
            </paragraph>
            <paragraph>
              <content styleCode="bold">MINOLIRA may cause serious side effects, including:</content>
            </paragraph>

                <content styleCode="bold">Harm to an unborn baby. See "What should I tell my healthcare provider before taking MINOLIRA?"</content>
              </item>
              <item>
                <content styleCode="bold">Permanent teeth discoloration.</content> MINOLIRA may permanently turn a baby or child's teeth yellow-gray-brown during tooth development. You should not use MINOLIRA during tooth development. Tooth development happens in the second and third trimesters of pregnancy, and from birth to 8 years of age.</item>
              <item>
                <content styleCode="bold">Slow bone growth.</content> MINOLIRA may slow bone growth in infants and children. Slow bone growth is reversible after stopping treatment with MINOLIRA.</item>
              <item>
                <content styleCode="bold">Diarrhea.</content> Diarrhea can happen with most antibiotics, including MINOLIRA. This diarrhea may be caused by an infection <content styleCode="italics">(Clostridium difficile)</content> in your intestines. Call your healthcare provider right away if you get watery or bloody stools.</item>
              <item>
                <content styleCode="bold">Liver problems.</content> MINOLIRA can cause liver problems that may lead to death. Stop taking MINOLIRA and call your healthcare provider right away if you get any of the following signs or symptoms of liver problems: 							<list listType="unordered" styleCode="circle">
                  <item>loss of appetite</item>
                  <item>tiredness</item>
                  <item>diarrhea</item>
                  <item>yellowing of your skin or the whites of your eyes</item>
                  <item>bleeding more easily than normal</item>
                  <item>confusion</item>
                  <item>sleepiness</item>
                </list>
              </item>
              <item>
                <content styleCode="bold">Increased pressure around the brain (intracranial hypertension).</content> This condition may lead to vision changes and permanent vision loss. You may be more likely to get intracranial hypertension if you are a female of childbearing potential and are overweight or have a history of intracranial hypertension. Stop taking MINOLIRA and tell your healthcare provider right away if you have blurred vision, double vision, vision loss, or headaches.</item>
              <item>Immune system reactions including a lupus-like syndrome, hepatitis, and inflammation of blood or lymph vessels (vasculitis). Call your healthcare provider right away if you get a fever, rash, joint pain, or body weakness.</item>
              <item>
                <content styleCode="bold">Serious skin or allergic reactions</content> that may affect parts of your body such as your liver, lungs, kidneys and heart. Sometimes these can lead to death. Stop taking MINOLIRA and go to the nearest hospital emergency room right away if you have any of the following signs or symptoms: 							<list listType="unordered" styleCode="circle">
                  <item>skin rash, hives, sores in your mouth, or your skin blisters and peels</item>
                  <item>swelling of your face, eyes, lips, tongue, or throat</item>
                  <item>trouble swallowing or breathing</item>

                  <item>fever, yellowing of the skin or the whites of your eyes, dark colored urine</item>
                  <item>pain on the right side of the stomach area (abdominal pain)</item>
                  <item>chest pain or abnormal heartbeats</item>
                  <item>swelling in your legs, ankles, and feet</item>
                  <item>darkening of your nails, skin, eyes, scars, teeth, and gums</item>

                    <content styleCode="bold">Discoloration (hyperpigmentation).</content> MINOLIRA can cause darkening of your skin, scars, teeth, gums, nails, and whites of your eyes.</item>

              <item>headache</item>
              <item>tiredness</item>
              <item>dizziness</item>
              <item>itching</item>

            <paragraph>MINOLIRA may cause fertility problems in males. This may affect your ability to father a child. Talk to your healthcare provider if you have concerns about fertility.</paragraph>
            <paragraph>Your healthcare provider may tell you to decrease your dose or completely stop taking MINOLIRA if you develop certain serious side effects during treatment with MINOLIRA.</paragraph>
            <paragraph>Your healthcare provider may do blood tests to check you for side effects during treatment with MINOLIRA.</paragraph>
